Here’s a fun one: https://iiif.elifesciences.org/lax/73346%2Felife-73346-fig2-figsupp8-v2.tif/full/1500,/0/default.jpg
“This is in line with the DNA nucleotide misincorporation profiles expected for the type of DNA library constructed (Seguin-Orlando et al., 2015), which was caused by the Q5 polymerase being unable to read through 5' uracils, thereby excluding the typical 5' excess of C-to-T.”

Do you happen to know/have/see the BAMs/raw damage patterns?
BAMs are here: https://www.ebi.ac.uk/ena/browser/view/PRJEB44527
Hi, made this 101-level sketch for 'Single stranded DNA Libraries'. If you like it I may try to improve 😅
*Thread Reply:* Keep going!
*Thread Reply:* However now I think about it... could we have some rights/permissions issues for using his likeness... do you know?
*Thread Reply:* But I really love the concept and idea 😄
*Thread Reply:* Haha have no clue about the rights issue...wish I thought about it before drawing for hours 😂
*Thread Reply:* This is where I took the pic from, and they say it's Fair Use. Would that be okay? https://www.wikiart.org/en/philippe-halsman/salvador-dali-1953
*Thread Reply:* I guess we won't make money off of it...
*Thread Reply:* I'll just put a big disclaimer 😄
*Thread Reply:* Could you send me the raw file(S)?
*Thread Reply:* Inkscape SVG would be perfect 😄
*Thread Reply:* As then I can re-export/convert as necessary (I also use it 🙂 )
*Thread Reply:* Here it is. Hope it won't cause trouble, apologies in advance 🥲
*Thread Reply:* You're based in Israel atm, right? (work wise)
*Thread Reply:* Eheheheh that's so cool, thank you @Nihan D Dagtas
*Thread Reply:* OPened perfectly 🙂
*Thread Reply:* (did you manually draw all the cells?!)
*Thread Reply:* Oooh actually, could you make a very slight tweak?
*Thread Reply:* Could you make the upper lip blue 😬
*Thread Reply:* Yes I can, and I'm in Tel Aviv yep
*Thread Reply:* Here with blue lip. I apparently ignored the contaminated sample completely 😂
*Thread Reply:* https://github.com/SPAAM-community/little-book-of-smiley-plots/pull/21
*Thread Reply:* All good @Nihan D Dagtas? and IC an merge it in 🙂
*Thread Reply:* Yes perfect, thanks for the opportunity. It was fun to make 🤗

Sorry if this is the wrong place to ask, but I've noticed that all my damage plots for single stranded libraries are not symmetric on the y-axis (figure attached). The 3' end consistently has a lower frequency across all my samples. I do see symmetry for my double stranded samples though. Could anyone explain why this is? The examples I have seen for single stranded libraries seem to be symmetric on the 5' and 3' ends.
*Thread Reply:* I would also like to know!
*Thread Reply:* I vaguely remember someone describing it at some point but I can't remember who
